It is a thing you do get penalized, depending on what position your dot is out of position the more the penalty will be.
rough example a te as a wr will be less penalize than say a ot playing qb

(took me an hour but I found this in my old notes)

OOP penalty/minimal penalized position subs

QB; Can use HB here for QB rushing plays

HB; Can use FB here. Also WR but only for passing if you don't have a Scat HB or FB (preferred).

FB; Can use HB for runs. Can use TE for blocking but needs to have good skill set. Slow, lack of agility, and low vision won't cut it.

TE; Can use OT for blocking or BFB. Scat FB works fine here as well. For some reason WR's don't run good routes from here.

WR;s Can use RTE's here (which makes no sense when WR's don't play well at TE) for passing... Scat backs as well. BTE's can block out here as well.

Centers; Only Guards can slide over without terrible damage to skills.

Guards; Centers or Tackles can play here.

Tackles; Guards or BTE's.

DT's; DE's only.

DE's; DT's or LB's.

LB's; DE's or Strong Safeties. Free Safeties don't seem to play well here. CB's neither but if you must in passing plays, it's not horrible.

CB's; Free Safeties are best. LB's if no other choice. However. You can put a Blitz LB (or SS) here and blitz the QB and it seems to work fine.

Strong Safety; LB's work best against the run. Free Safeties against the pass.

Free Safety; Strong Safety against the run, CB's against the pass.

Special note; In Special Teams formations OOP penalties seem to have no effect at all. Use Defensive players in offensive formations and vise versa, especially when punting.

As you can see this was copied from some time ago... back before I did ST coach work and was just a total newb at doing OC and DC for Peewee leagues. I believe I got this from someone who was teaching me at the time and was a damn good coach and mentor so I've trusted it over the years (decades?). Hope it helps you as it helped me.

The cynical answer is it has to do more with flex cost and game balance than actual logic.

Like a pass rush DE is way better than blitz LB because of SAs. So there has to be a penalty for playing a DE at LB or else everyone would do it.

If receiving TEs didn't have a significant penalty at WR, why would anyone ever buy a WR? TEs have the better Majors and Minors, acceptable SAs, and they get to be taller and heavier.

TL;dr is the penalties are significant enough to be avoided if at all possible, unless the sheer superiority of the dot is enough to overcome the penalties. Like a 280 pound blocking TE with great SAs and attributes as a blocking WR.
