What exactly do Play Priorities do under the playbook? Do they choose which plays they will most call? Someone please explain.

First your tactics have to dictate which pool of plays you are pulling from. For offense this is your different pools with runs and your different pools of passes. For defense it goes off of which formation the opposing offense has in and then what you tactics are pointing to whether it is a specific play distance tag (short, medium, long) along with your other selections like man or zone along with blitz. Once a pool of plays has been selected if there is more than one play which can fire in that tactical situation the priorities come in to play and a 5 priority play has a greater chance to fire than a play in the same pool with less than the 5.
