I may need to make a few guys for inactive players, do I have to day 48 to make an exp rookie?

I think they cut it off on Day 47 last season but I could be wrong. Its important to note, any player built now will not count towards a SS and if you retire players on Day 48 they won't count either.

Just to be clear and include the last bit-
"Its important to note, any player built now will not count towards a SS" (this season). If you keep them they'll count the next season when you get them to the next tier.

Just be safe and make them now. That way they'll also get a few games of chem. Plus, those inactives haven't spent all of their SP so they're probably near the same level as a fully spent player with 0 chem.

This is the way to go - if they've been innactive since the last * point skill boost (basically, a week or so), a new player will be better most of the time. Plus, rosters lock in the playoffs, so those inactives just get worse, while your new guys again get that chem boost.
